Summary:One of the most important complication that may develop due to Incorrect handling with diabetes mellitus is foot ulcer that can lead to resection part of leg. Several study show that Oxidative stress [OS] paly a key in the development of diabetic foot ulcer and other complication associated with diabetes, the research indicates the healing of diabetic wound was highly affected by the decrease of nitric oxide. The polymorphism of Endothelial Cell Nitric Oxide Synthase 3 [eNOS] Glu298Asp and their association with the development of diabetic foot ulcer and oxidative stress level in Iraqi patients with type two diabetis mellitus was the aim of this study. Methods In this case-control study, 50 patients with type 2 diabetes and diabetic foot ulcer [DFU], 50 patients with type 2 diabetes without DFU and 50 healthy control group were recruited. We used restriction fragment length polymorphism [RFLP-PCR] technique for the genotyping of eNOS Glu298Asp polymorphism in exon 7. In this study the measurement of ferric-reducing and thiobarbituric reactive level was used to determination the ability of plasma as the potential markers of OS. Results study shows that the frequency T allele in control was significantly elevated than patients groups as well as there is a significant differences between patients and healthy group in the frequencies of eNOS Glu298Asp polymorphism. The results show that the level of TBARS & FRAP levels were significantly different among the several genotypes. Conclusions The study conclude that the T allele in eNOS gene may give a protection from development foot ulcer in diabetic patients.
