Summary:Indonesia is one of the countries with high biodiversity of freshwater fishes. One of the rivers with high freshwater fish diversity is Arut River located in West Kotawaringin, Central Kalimantan. Freshwater fish which is commonly found in the Arut River is striped snakehead. The fish is popular to be consumed by the community due to its delicate and its high nutrient content. However, the research on the genetic characterization of the striped snakehead from Arut River by applying the COI mitochondrial gene has not been conducted. By this reason, the aim of this research was to analyze the genetic characterization of the striped snakehead from Arut River by using the COI mitochondrial gene. The research applied a PCR method with primer F2 and R2. The result exhibited that the five samples of the striped snakehead investigated in this study had 2 haplotypes with 1 variable site without parsimony site. The haplotype diversity and nucleotide diversity were 0.400±0.237 and 0.00068± 0.0004 respectively with a genetic distance of approximately 0.08%. This finding indicates that intrapopulation genetic variation of the fish species is low. In addition, both haplotypes formed on the striped snakehead investigated in this study was different from the striped snakehead from other regions in Indonesia and Southeast Asia, so it can be used as a molecular marker of the striped snakehead from Arut River.
